Enzymatic oxidation of nicotine to nicotine 1'(5') iminium ion. A newly discovered intermediate in the metabolism of nicotine.

چکیده
Oxidation of nicotine in the presence of rabbit liver 10,000 x g supernatant, NADPH, and cyanide results in the production of 5’-cyanonicotine. NaBDl reduction of the reaction mixture from a similar incubation of nicotine, NADPH, and supernatant results in the production of deuteronicotine having a single deuterium in the pyrrolidine ring. Oxidation of nicotine to cotinine under 1802 did not lead to incorporation of I80 in the cotinine produced. These results are explained by the formation of nicotine AlfC6’) iminium ion as an intermediate in the degradation of nicotine. The formation of the iminium ion is catalyzed by an enzyme system having the properties of a mixed function oxidase. The reaction requires NADPH and O2 and is inhibited by carbon monoxide. It is proposed the nicotine A1’(6’) iminium ion is formed by loss of water from 5’-hydroxy nicotine. The iminium ion formed from nicotine represents a nicotine analogue with a markedly altered chemical reactivity and must therefore be considered in studies on the physiological action of nicotine.

In smokers, the primary pathway of nicotine metabolism is P450 2A6-catalyzed 5'-oxidation. The nicotine Delta(5'(1'))-iminium ion product of this reaction is further metabolized to cotinine by aldehyde oxidase. Previous investigators have reported kinetic parameters for cotinine formation using human liver cytosol as a source of aldehyde oxidase. Using [5-(3)H]nicotine and radioflow high-perfor...
متن کاملCYP2A6 genetic polymorphism and its relation to risk of smoking dependence in male Iranians
Introduction: Nicotine is the psychoactive substance responsible for establishing and maintaining smoking dependence. CYP2A6 is the primary enzyme that inactivates nicotine to cotinine .Genetic variation in CYP2A6 accounts for some of the inter-individual variability in nicotine metabolism and has been indicated to influence smoking behavior and dependence. Therefore, the aim of this study was ...

Nicotine is the major addictive agent in tobacco. The primary catalyst of nicotine metabolism in humans is CYP2A6. However, the closely related enzyme CYP2A13 is a somewhat better catalyst. CYP2A13 is an extrahepatic enzyme that is an excellent catalyst of the metabolic activation of the tobacco-specific carcinogen 4-(methylnitrosamine)-1-(3-pyridyl)-1-butanone (NNK). Here we report that both C...
متن کاملAscorbic acid inhibits nicotine-induced behavioral sensitization in male mice
Repeated administration of nicotine causes incentive and behavioral sensitization in animals. Ascorbic acid administration inhibits some effects of nicotine. In the present study, the effect of ascorbic acid administration on nicotine-induced behavioral sensitization in Male Swiss-Webster mice (20-25 g) was investigated. Animals were injected with nicotine (0.25 mg/kg, i.p.) once daily for seve...
